A last-ditch drive over the line scored by No 8 Ethan Clarke and converted by fly-half Jake Rodgers gave Tynedale a crucial win.
It lifted them above Sheffield Tigers into third in the table and showed their promotion credentials.
Clarke's try was the third for the Northumberland side after earlier scores by centre Jonny Burden and prop Mike Hughes while the hoodoo of never beating Tynedale continues for the Tigers.
Sheffield had a chance to steal the win in the final play after Tynedale defenders gave away a penalty at the ruck for holding on, but fly-half Mark Ireland's 40 metre penalty kick went short.
Tynedale's director of rugby Ben Woods said "It was a case of two top teams going at it and Sheffield Tigers are really good.
"You can see why they are in such a good position, and either team would be disappointed to lose. So, to come out on the right end, is pleasing. It was just a really good game of rugby.
"We're really pleased with our line-out including stealing several of theirs.
"The boys make it a big aspect for us and it makes it difficult for opposition. In the first five minutes they had five goes at it and we could have been 14 or more points down.
"We made a few good line breaks, but their back-three were fantastic in covering and bring a nuisance at the breakdown.
"We've edged a really tight game with a lot of composure"
With both teams on four-game unbeaten runs,the game was end-to-end and full of endeavour from kick-off but both defensive lines proved dominant.
Tigers pinned themselves into Tynedale's 22 but lost three crucial attacking line-outs out of four in the first 15 minutes.
They struggled throughout the game, losing seven out of 18 by the final whistle. Centre Tom Hart looked odds-on, but for a try-saving intercept by Tynedale blindside Jonny Cousin.
Tynedale's catch-and-drive came close and they won two more penalties in Tiger's territory. However, they went for a try rather than kick for points, which proved costly, Prop Hughes' try was chalked off for a double mvement.
A 20-metre catch-and-drive finished by Tigers openside Jack Redfern-Brown broke the deadlock but Tynedale narrowed the score when Burden intercepted a Hart pass and ran in 30 metres.
Tigers wing Peter Swatkins and centre jamie Broadley combined for the latter to score straight after half-time but a sloppy lineout allowed Tynedale's Hughes to touch down.
The tight affair continued until Clarke's winning try making up for losing to league leaders Fylde five weeks ago.
Tigers player-coach Jamie Broadley said "We're really frustrated, to lose by a point in a game where you give away an intercepted try and have a brain-lapse on our own five-metre line for a questionable try.
"Our performance was really good for 98 percent but Tynedale are such a good side they capitalised on that two per cent, got their scores and that was the difference."
